اهداف: زرين گياه داراي بيش از 60 گونه است و عمدتا در مناطق معتدل آسيا و اروپا یافت می شود. یکی از این گونه ها که بادرنجبویه دنایی نام دارد در بسیاری از نقاط جهان دارای خواص دارویی و اجزای فعال است. با وجود آن که یک گیاه بومی وحشی مهم است، اما در حال حاضر، این گیاه در ایران در معرض خطر قرار دارد. علاوه بر این، در مورد اهمیت خواص دارویی و اجزای فعال بادرنجبویه دنایی، در میان مردم ایران اطلاعات کمی وجود دارد. بر این اساس، بررسی سیستماتیک مطالعات مربوط به خواص دارویی و اجزای فعال و اهمیت آن در سلامت انسان و حیوان انجام شد و شکاف های دانش موجود شناسایی شده اند. آیتم های گزارش شده مورد نظر برای بررسی های سیستماتیک و دستورالعمل های متا آنالیز (PRISMA) در جست و جو برای مقالات منتشر شده در مورد خواص دارویی و اجزای فعال زرین گیاه و اهمیت آن برای انسان و حیوان در ایران مورد استفاده قرار گرفتند. این جست و جو محدود به مقالات علمی پایگاه داده های محبوب و موتور های جست و جو مانند PubMed، Web of Science، Google Scholar، Science Direct، SpringerLink و Scopus بود. جست وجو محدود به مجلات علمی، کتاب ها و فصل های کتاب بود که بر خواص دارویی زرین گیاه در ایران در طول سال های 1970 تا 2018 متمرکز بودند. به طور کلی، 1158 مقاله علمی از پایگاه های مختلف استخراج شدند که از این میان 38 مقاله معیارهای جست و جو را برآورده ساختند و برای این بررسی مجاز بودند. این مطالعات فقط در 9 استان از 31 استان ایران انجام شدند و بخش زیادی از آن ها در استان اصفهان، مرکز ایران صورت گرفتند. مطالعات انجام شده نشان دادند که همه بخش های گیاه (ریشه ، بخش های هوایی، گل ها، برگ ها) دارای اجزای فعال هستند. روغن های اساسی و بخش های فعال گیاهان، اجزای اصلی مورد مطالعه بودند. با این وجود، رایج ترین اجزای گزارش شده زانتومیکرول، لیمونن، لووتولین، ژرانیل، آپی ژنین و کالیکوفترین بودند. تعدادی از خواص دارویی گزارش شده میان آن ها عبارت اند از آنتی اکسیدن ها، آنتی باکتری ها، ضد سرطان ها، آنتی هایپرلیپیدمیک، صد اسپاسم و ضد اثرات سیتوتوکسیک. علاوه براین، گزارش شد که این گیاه درمانی برای درد التهابی، سردرد، احتقان، اختلالات کبدی، زخم، تب، درد کلیوی، سوء هاضمه، درد معده، درد شکم، درد مفاصل، اسپاسم عضله، احتقان ، التهاب و اثرات التهاب زخم است. نتیجه گیری: این مطالعه نشان داده است که زرین گیاه یک گیاه دارویی مهم با چند جزء فعال و قابلیت بالا برای حفاظت از سلامت انسان و حیوان در ایران است. با این حال، استفاده بیش از حد از زرین گیاه، وجود آن را در معرض خطر قرار می دهد. با این وجود، مطالعات بیش تری باید در کشور انجام شوند.

نتیجه گیری

این مطالعه نشان داد که بادرنجبویه دنایی دارای اجزای مفیدی است که می توانند برای توسعه دارو مورد استفاده قرار گیرند. در حال حاضر، شواهد حاکی از آن است که گیاهان دارویی دارای خواص داریی با عوارض جانبی همراه هستند. نتیجه این مطالعه نشان دهنده اثر سیتوتوکسی نسبتا قوی عصاره متانول بادرنجبویه دنایی و محتوای بالای فنل ها و فلاونوئیدها در این عصاره ها است. بر اساس این نتایج، بادرنجبویه دنایی می تواند یک گزینه مناسب برای طراحی و تولید دارو های ضد سرطان باشد. با این حال، همانطور که از این بررسی پیدا است، خواص دارویی بادرنجبویه دنایی بر تعدادی از این بیماری ها هر دو در انسان و حیوان مانند اثرات هر دو در انسان و حیوانات مانند ضد ویروسی، ضد قارچی، ضد سمیت، فعالیت عصبی، ایمن سازی و فعالیت های ترویجی به خوبی مورد ارزیابی قرار نگرفته اند. علاوه بر این، تحقیقات مبتنی بر ایمنی سمیت، به ویژه در مقادیر LD50 مناسب از طریق آزمایشات آزمایشگاهی و مطالعات بالینی مورد نیاز می باشند. اگر چه برخی از محصولات گیاهی بادرنجبویه دنایی به طور سنتی توسط افراد محلی در بخش هایی از ایران تهیه شده اند، اما قابلیت آن برای بهره برداری بهتر در سراسر ایران و سراسر جهان مورد استفاده قرار نگرفته است. با توجه به اینکه بادرنجبویه دنایی یک گونه در معرض خطر در ایران است، برای حفاظت از گیاه و بنابراین جلوگیری از حذف احتمالی آن به تلاش بیش تری نیاز است.

Objectives. Dracocephalum has over 60 species and is found mainly in the temperate regions of Asia and Europe. One of these species, i.e., Dracocephalum kotschyi Boiss, is known to have a number of medicinal properties and active ingredients in many parts of the world. Despite being an endemic wild-fowering plant of great importance, the plant is currently considered endangered in Iran. Besides, there is paucity of information on the signifcance of the medicinal properties and active constituents of D. kotschyi among the Iranian people. On that account a systematic review of studies reporting on the medicinal properties and active ingredients and its signifcance to human and animal health was conducted and the existing knowledge gaps were identifed. Methods. Te Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines were used in the search for published articles on medicinal properties and active ingredients of D. kotschyi and its signifcance on humans and animals in Iran. Te search was confned to scientifc articles from repositories of popular data bases and search engines among them PubMed, Web of Science, Google Scholar, Science Direct, SpringerLink, and Scopus. Te search narrowed down on scientifc journals, books, and book chapters focusing on the medicinal properties of D. kotschyi in Iran for the period between 1970 and 2018. Results. A total of 1158 scientifc articles were sourced from the various databases, out of which 38 met the search criteria and qualifed for this review. Te studies were conducted in only 9 of the 31 provinces of Iran, with a large proportion in Isfahan province, central Iran. Te studies showed that all plant parts (roots, aerial parts, fowers, and leaves) had active constituents. Essential oils and aerial plant parts were the main components studied. Nevertheless, the most frequently reported constituents were xanthomicrol, limonene, luteolin, geranial, apigenin, and calycopterin. A number of medicinal properties were reported among them antioxidant, antibacterial, anticancerous, antinociceptive, antihyperlipidemic, antispasmodic, cytotoxic, and immunomodulatory efects. Te plant was also reported to be a remedy for infammatory pain, headaches, congestion, liver disorders, ulcer, fever, renal pain, dyspepsia, stomach ache, abdominal pain, joints pains, muscle spasm, congestion, bloating, and wound healing efects, among others. Conclusion. Tis review has shown that D. kotschyi is an important medicinal plant with a large number of active constituents and great potential to safeguard human and animal health in Iran. However, over utilization of the D. kotschyi plant is already endangering its existence. Nevertheless, more studies need be conducted across the country

5- Conclusion

Tis review has noted that D. kotschyi has several benefcial ingredients that can be exploited for drug development. Currently, surmounting evidence suggest that medicinal plants have a number of medicinal properties ofen with minimal side efects. Results of this review further showed a relatively strong cytotoxic efect of methanol extract of D. kotschyi aerial parts and the high content of phenols and favonoids in these extracts. Based on these results, D. kotschyi can be a suitable choice for designing and manufacturing anticancer drugs. However, as is evident from this review, the medicinal properties of D. kotschyi on a number of diseases, both in human and in animals such as antiviral, antifungal, antinephrotoxicity, neuroprotective, immunomodulatory, and growth promoting activities, have not been well evaluated. Additionally, research based on safety and toxicity, especially on the appropriate LD50 values through laboratory tests, and clinical studies are needed. Tough some herbal products of D. kotschyi have been prepared and used traditionally by the locals in parts of Iran, its potential has not been fully exploited for better utilization across Iran and around the worldwide. Given that D. kotschyi is an endangered species in Iran, there is need for concerted efort to conserve and protect the plant and therefore subvert its eventual elimination.

چکیده

قرارگیری در معرض حشره‌کش ممکن است آثار میان‌نسلی و چندنسلی را در جمعیت‌ها ایجاد نماید، اما مکانیسم‌های مولکولی این تغییرات تا حد زیادی ناشناخته باقی‌مانده است. اکثر مطالعات بر روی مکانیسم‌های میان‌نسلی یا چند‌نسلی متمرکز شده‌اند اما از جنبه‌های مقایسه‌ای آنها غافل مانده‌اند. این مطالعه بررسی کرد که آیا حشره‌کش پیرتیروئید اتوفن‌پراکس (فرمولاسیون EC 30 تربون®) آثار میان‌نسلی و یا چند‌نسلی بر روی بقا و تولید مثل فولسومیا کاندیدا(کولمبولا) دارد یا خیر. فعال سازی ژن‌های وابسته به تنش برای کشف اینکه اتوفن‌پراکس بیان ژن‌های وابسته به تولید مثل را در تیمارهای میان و چند‌نسلی تغییر می‌دهد یا خیر، بررسی گردید. یک مطالعه‌ی آزمایشگاهی برای سه نسل با غلظت‌های حشره‌کش در خاک لوفا2.2 انجام گرفت. در تیمار چند نسلی، تنها نسل والدین (P) مورد بررسی قرار گرفت، اما برای نسل‌های بعدی چنین نبود. در تیمار چندنسلی، هر سه نسل در یک روش مشابه در معرض حشره‌کش قرار گرفتند. قرارگیری در معرض حشره‌کش موجب کاهش آثار تولیدمثل در طول نسل‌ها می‌شود و پیشنهاد می‌شود که فولسومیا کاندیدا قادر به سازگاری با افزایش سطح غلظت اتوفن‌پراکس در طولانی مدت در طی چندین نسل است. در تیمار میان‌نسلی، بیان پروتئین شوک حرارتی 70 در بالادست و سیتوکروم اکسیداز 1v4N6 در پایین دست در دوز وابسته به نسل F2 تنظیم گردید. این یافته‌ها احتمال وراثت اپی‌ژنتیکی آثار حشره‌کش بر والدین را بالا می‌برد. بعلاوه، بیان سیتوکروم اکسیداز 1v4N6 برخلاف تیمارهای میان و چند‌نسلی تنظیم شد. نتایج ما توجهات را به سمت اختلاف پاسخ‌ها در سطح مولکولی فلوسمیا کاندیدا به قرارگیری در معرض اتوفن‌پراکس میان و چند‌نسلی جلب می‌نماید.

5- نتیجه‌گیری

حشره‌کش اتوفن‌پراکس، در فرمولاسیون تربون ® 30EC، نشان داد که در دو نسل میان نسلی و چندنسلی بر روی فلوسومیا کاندیدا اثر داشته است. در تیمار میان نسلی، اثرات روی زنده‌ها و تولیدمثلی در نسل اول مشاهده شد، اما این یافته ممکن است ناشی از قرارگیری مستقیم نابالغ‌های نسل اول در معرض حشره‌کش باشد. اثرات تراریخته‌ی ظاهری اتوفن‌پراکس در رونویسی پروتئین شوک گرمایی 70 و سیتوکروم اکسیداز 1v4N6 در نسل دوم بدست آمد. بیان پروتئین شوک گرمایی70 در بالادست و سیتوکروم اکسیداز 1v4N6در پایین دست به صورت وابسته به دوز و بدون اثر مستقیم آفت‌کش تنظیم شدد. مطالعه‌ی بیشتر مکانیسم‌های مولکولی در پس تغییرات بیان سیتوکروم اکسیداز 1v4N6و پروتئین شوک گرمایی70 نیاز است. اثرات چندنسلی به عنوان اثر کاهشی روی تولیدمثل (تعداد نابالغ‌ها) در نسل اول و دوم بیان شده است. پاسخ‌های سلولی به اتوفن‌پراکس در تیمارهای میان نسلی و چندنسلی ظاهر شد و حساسیت بالایی را نشان داد. مقدار RNAی بدست آمده در تمام تیمارها به طور وابسته به دوز کاهش یافت(به جز در نسل میان نسلی2). که حاکی از ضعف سلامتی کولمبولانس‌های در معرض اتوفن‌پراکس است. الگوهای ترانسکریپتومی ژن‌های تنش می‌تواند حاکی از تنش اتوفن‌پراکس در جمعیت باشد. ژن پروتئین شوک گرمایی70 نشان دهنده‌ی تنش عمومی است اما ژن سیتوکروم اکسیداز 1v4N6در ارتباط با سم‌زدایی زنوبیوتیک‌های دارای پیوند استری است. ژن‌های فعال شده در تیمار میان نسلی و کاهش مهار تولیدمثل در تیمار چندنسلی مشابه به مرحله‌ی اول مقاومت یا سازگاری است.

Abstract

Insecticide exposure may cause both transgenerational and multigenerational effects on populations, but the molecular mechanisms of these changes remain largely unclear. Many studies have focused on either transgenerational or multigenerational mechanisms but did neglect the comparative aspects. This study assessed whether the pyrethroid insecticide etofenprox (formulation Trebon® 30 EC) shows transgenerational and/or multigenerational effects on the survival and reproduction of Folsomia candida (Collembola). The activation of stress-related genes was studied to detect whether etofenprox modifies the expression of reproduction-associated genes in trans- and multigenerational treatments. A laboratory study was carried out for three generations with five insecticide concentrations in LUFA 2.2 soil. In the transgenerational treatment, only the parent generation (P) was exposed, but the subsequent generations were not. In the multigenerational treatment, all three generations were exposed to the insecticide in the same manner. Multigenerational exposure resulted in reduced reproduction effects over generations, suggesting that F. candida is capable of acclimating to enhanced concentration levels of etofenprox during prolonged exposure over multiple generations. In the transgenerational treatment, the heat shock protein 70 was up-regulated and cytochrome oxidase 6N4v1 expression down-regulated in a dose-dependent manner in the F2 generation. This finding raises the possibility of the epigenetic inheritance of insecticide impacts on parents. Furthermore, CYP6N4v1 expression was oppositely regulated in the trans- and multigenerational treatments. Our results draw attention to the differences in molecular level responses of F. candida to trans- and multigenerational etofenprox exposure.

5- Conclusions

The insecticide etofenprox, in the formulation Trebon® 30 EC, showed both transgenerational and multigenerational effects on F. candida. In the transgenerational treatment, effects on survival and reproduction were observed in the F1 generation, but this finding may be due to direct exposure of the F1 juveniles to the insecticide. Apparent transgenerational effects of etofenprox on the transcription of HSP70 and CYP6N4v1 were found in the F2 generation. The HSP70 expression was upregulated and the CYP6N4v1 down-regulated in a dose-related manner without direct pesticide exposure. Further study of the molecular mechanisms behind the observed HSP70 and CYP6N4v1 expression changes is required. Multigenerational effects are expressed as a reduced effect on reproduction (juvenile numbers) at F1 and F2 generations. Cellular responses to etofenprox appeared both in transand multigenerational treatments, and showed high sensitivity. The obtained amount of RNA was decreased in all treatments in a doserelated manner (except in TF2 generation), suggesting poor health of the etofenprox-exposed collembolans. The stress-connected genes reacted with a dose-related increase to etofenprox exposure, which suggests that the population could be more resistant to further insecticide exposure or other environmental stress factors. The transcriptomic patterns of the stress genes could indicate an etofenprox stress of the population. The HSP70 gene indicates general stress, but the CYP6N4v1 gene is connected with the detoxification of ester bond-containing xenobiotics. The activated genes in the transgenerational treatment and the reduced inhibition of reproduction in the multigenerational treatment both resemble an early stage of resistance or acclimation.

چکیده

     آزمايشي با هدف ارزيابي تاثير افشانه هاي كلسيم حاوي نانوذرات (نانو كلسيم) و كلسيم کلراید (CaCl2.2H2O) بر روی برگ و مشاهده عملكرد و كيفيت گونه زراعی انار اردستانی در طی دو سال متوالی 2014 و 2015 انجام شد. کود نانو کلسیم در غلظت های 25/0 و50/0 گرم کلسیم L-1 افشانده شد و CaCl2.2H2O در غلظت های 1 و 2 درصد (2/73 و5/45 g Ca L−1) مورد استفاده قرار گرفت. نحوه ی تیمار شدن به این صورت بود که در ابتدا در زمان شکوفه کامل و برای بار دوم در یک ماه بعد این کار انجام می شود. افشانه های کلسیم اثر معنی داری بر عملکرد، تعداد میوه در هر درخت و وزن متوسط میوه نداشته است، در حالی که باعث افزایش طول میوه، فقط در مورد تیمار 1٪ CaCl2 در فصل اول شده است. درختان تیمار نشده در باغ نسبتاً تحت تاثیرترک خوردگی میوه قرار گرفتند، که 6-7٪ از میوه ها تاثیر پذیرفتند. تیمار برگ ها با محلول افشانه ای کلسیم با کود نانو کلسیم در غلظت 5/0 گرم Ca L−1 و 1 % Ca L−1 ( در هر دو فصل) و همچنین 2 % Ca L−1 (فقط در فصل دوم) در مقایسه با گروه شاهد، ترک خوردگی میوه را به میزان قابل توجهی کاهش داد و در نتیجه باعث افزایش عملکرد میوه در بازار شد. افشاندن 1 % Ca L−1 باعث افزایش 6 یا 7 % TSS ، فقط در فصل دوم شده است. علاوه بر این، کود کلسیم 50/0 گرم Ca L−1 منجر به کاهش جزئی (تقریبا 1٪) در کل فنل ها فقط در فصل اول شد. سایر خصوصیات شیمیایی، از جمله اسیدیته قابل اندازه گیری، بلوغ میوه، قند کل، فعالیت آنتی اکسیدانی و محتوای آنتوسیانین کل در لقاح کلسیم تحت تأثیر قرار نگرفت. تجزیه و تحلیل برگ نشان می دهد که تیمارهای برگ باعث افزایش غلظت کلسیم برگ در فصل اول، به استثنای دوز کم کود نانو، شده است؛ در حالی که غلظت برگ های N ، P ، K ، آهن، روی و منگنز بی تاثیر بودند. به طور خلاصه، باروری با غلظت کم (50/0 g CaCl2) کلسیم در فرمول نانو کلسیم منجر به کاهش مشابه در ترک خوردگی میوه انار نسبت به موارد به دست آمده با دوزهای بالاتر CaCl2 شد ( 73/2 و 45/5 g CaCl2).

مواد و روش ها

محل آزمایشی، مواد گیاهی و تیمارها

     این آزمایش در دو فصل 2014 و 2015 در یک باغ انار تجاری انجام شد. باغ در قسمت مرکزی استان خراسان رضوی در شمال شرقی ایران واقع شده است (Tous Dasht; lat. 35° 1′ 24.33” N, long. 58° 50′ 19.61” E, altitude 967 m) و خاک بعد از تکه تکه شدن به صورت خاک گلدانی درشت، مخلوط می باشد ( 64% ماسه ، 12٪ خاک رس و 24٪ سیلت). با pH برابر 8.08 در آب و EC 9.4 dS m-1، (EC: کمیسیون اروپایی). منطقه خشک است و 248 میلی متر متوسط بارندگی در آن سال است و میانگین سالانه °C 8 /14 درجه سانتی گراد است. درختان هشت ساله با سه تنه، تقریباً 5/2 -3 متر ارتفاع داشتند. درختان در ردیف های منظم و در فاصله 5 ×3 متر (667 درخت در هکتار) کاشته شدند و توسط یک سیستم آبیاری قطره ای آبیاری شدنددر باغ مورد بررسی، ترک خوردگی میوه حتی با وجود استفاده از سیستم آبیاری قطره ای و برداشت قبل از سرد شدن هوا و در پایان فصل وجود داشت.

     دو منبع کلسیم مورد استفاده قرار گرفت، یک “کود نانو کلات شده ” Ca (250گرم Ca L-1 ؛ پس از آن به نام nano-Ca ) و. CaCl2.2H2O آزمايش بر اساس طرح بلوك كاملاً تصادفي با پنج تيمارControl −no Ca، nano-Ca1 ، nano-Ca2 ، CaCl2 1٪، CaCl2 2٪ و چهار تكرار در هر تيمار انجام شد. چهار ردیف درخت انار انتخاب شد و در هر ردیف یک درخت با هر 5 تیمار اسپری شد. کود نانو کلسیم حاوی نانوذرات (ترکیب ثابت و مورد تایید، اندازه متوسط 50 نانومتر، دامنه 23-80 نانومتر) است و در غلظت های 25/0 و 50/0 گرم کلسیم L-1 برای استفاده در اسپری ها و کودهای افشانه ای مورد استفاده قرار گرفت (nano-Ca1 و nano-Ca2 به ترتیب؛ دوز توصیه شده از طرف تولید کننده 50/0 می باشد). از طرف دیگر، CaCl2 در غلظتهای 73/2 و 5/5 گرم از کلسیم L-1 (1 % و2 %CaCl2 · 2H2O ، به ترتیب)دوزها قابل مقایسه با دوزهایی هستند که در مطالعات قبلی کود پاشی محلول روی برگ در گیاه انار مورد استفاده قرار گرفت (and 4%; Ramezanian et al., 2009). محلول کود به وسیله ی رقیق کردن nano-Ca محصول مایع تجاری و یا با CaCl2·2H2O توسط آب زیرزمینی موجود در چاه های موجود در باغ تهیه شده است. درختان دو بار در هر فصل اسپری می شوند، اولین بار در زمان شکوفه کامل، در 12 مه 2014 و 26 آوریل 2015 و بار دوم یک ماه پس از اولین اسپری می باشد. در کلیه موارد با 5/3 لیتر در هر درخت (تا مرطوب شدن کامل شاخ و برگ ؛ کل دوز به کار رفته با کودهای nano-Ca و CaCl2 تقریبا 3/1 یا 7/2 گرم کلسیم برای هر درخت، 5/14 یا 9/28 گرم کلسیم برای هر درخت، به ترتیب استفاده شد). برگ ها از قسمت میانی شاخه های میوه برای نمونه برداری انتخاب شد (100 برگ از سه تنه در اطراف تاج گل در هر درخت) در 11 اوت در فصل اول و در 6 اوت در فصل دوم نمونه این کار انجام شد. میوه ها در 22 اکتبر در فصل اول و 14 اکتبر در فصل دوم برداشت شدند، که تاریخ برداشت آن بر اساس شکل کلی و ظاهری میوه و خواص شیمیایی میوه است.

اندازه گیری گیاه

     غلظت مواد مغذی و ریز مغذی های موجود در برگ انار در آزمایشگاه ایران در فصل اول و در آزمایشگاه اسپانیا در فصل دوم اندازه گیری شد. در ایران نمونه هایی بر اساس تحقیقات Chapman و Pratt (1961) آماده شدند؛ و کل ، N ، P ، K  و Ca به ترتیب با استفاده از روش Kjeldhal، اسپکتروفتومتری، فتومتر و جذب تابش انجام شد. محتوای آهن، روی و منگنز با استفاده از طیف سنجی جذب اتمی تابش اسپکتروفوتومتری اندازه گیری شد. در آزمایشگاه اسپانیا با استفاده از دستگاه مایکروویو مورد برسی قرار گرفت و برای آهن، منگنز، روی و کلسیم با استفاده از طیف سنجی جذب اتمی نور اسپکتروفوتومتری و برای K با استفاده از طیف سنجی انتشار نور مورد بررسی قرار گرفت (Carrasco-Gil et al., 2016). سه تکرار در هر درمان و در هر سال انجام شد.

چکیده

هدف ما مطالعه ي اثرات گليفوسات، كشت و سابقه آن بر كلونيزاسيون ميكوريزا و رشد گياهان هدف (علف هاي هرز) و غير هدف (محصولات) بود. گليفوسات، رايج ترين كشنده حشرات در جهان، يك آنزيم موجود در گياهان و ميكروب ها را مهار مي كند. ما اثرات گليفوسات اعمال شده در پاییز قبلی را بر رشد یک علف دائمی، بیدگیاه (گیاه هدف) و یک علف، فتوک ( گیاه غیر هدف) و کلونیزاسیون ریشه (AMF) گیاه قارچی میکوریزا آربوسکولار بررسی کردیم. گیاهان غیر هدف در بهار سال بعد کاشته شدند. علاوه بر این، بررسی شد که آیا اثرات گلیفوسات به کشت یا ویژگی های خاک تنظیم شده با سابقه طولانی کشت علف همزیست اندوفیت بستگی دارند یا نه (علف E+). کلونیزاسیون ریشه AMF ، ایجاد و رشد گیاه، بقایای گلیفوسات در گیاهان، و شیمی خاک مورد سنجش قرار گرفتند. گلیفوسات، کلونیزاسیون میکوریزا و رشد علف های هدف و غیر هدف را کاهش داد. با توجه به سابقه کشت علف E+ ، میزان کاهش به کشت و خواص خاک بستگی داشت. ما در فصل رشد، پس از اعمال گلیفوسات، از بقایای گلیفوسات در علف ها و گیاهان استفاده کردیم. بقایا در گیاهانی که در گلدان های بدون خاک رشد می کردند، بیش تر از همگونه های موجود در گلدان های دارای خاک بودند. این نتایج نشان دهنده اثرات منفی گلیفوسات بر ارگانیسم های غیر هدف در محیط های کشاورزی و اکوسیستم های چمنزار هستند.

نتیجه گیری

نتایج ما نشان می دهند که اثرات شیوه های کشاورزی شیمیایی و مکانیکی بر خدمات اکوسیستم می توانند فراتر از سال باشند. در این جا نشان دادیم که گلیفوسات، AMF مفید گیاه در علف های هدف و غیر هدف را به طور منفی تحت تاثیر قرار می دهد. این نتایج توجه ما را به سوی درک جامع تر اثرات گلیفوسات بر میکروب های مرتبط با گیاه و جوامع میکروبی موجود در خاک ها جلب می کنند. به عنوان مثال، مشخص شده است که میکروب های خاک در تجزیه گلیفوسات نقش مهمی ایفا می کنند (آیانسینا و اوسو 2006) و بنابراین، تغییرات مربوط به فرکانس و ترکیب جامعه آن ها می توانند مقدار گلیفوسات موجود در گیاهان و علف ها را تحت تاثیر قرار دهند. علاوه بر عوامل حیاتی، محیط فیزیکی نیز در تعیین کنش های گلیفوسات در خاک اهمیت دارد. بسته به ریزجانداران اولیه خاک، گلیفوسات می تواند نتیجه ترکیب میکروبی مربوط به تجزیه گلیفوسات و علف ها را به طور متفاوتی تغییر دهد. به طور خاص، در F. pratensis غیر هدف، کلونیزاسیون AMF به صورت تعاملی با عوامل حیاتی مانند قارچ های سیمبیوتیک پوشش گیاهی قبلی و عوامل فیزیکی مانند تکنیک های کشت، کاهش می یابد. موضوع اصلی در حفظ خاک های حاصلخیز برای تولید پایدار محصولات ارائه محیط مطلوب برای خاک و میکروب های مفید گیاهی است.

Abstract

Our aim was to study the effects of glyphosate, tilling practice and cultivation history on mycorrhizal colonization and growth of target (weeds) and non-target (crops) plants. Glyphosate, the world’s most widely used pesticide, inhibits an enzyme found in plants but also in microbes. We examined the effects of glyphosate treatment applied in the preceding fall on growth of a perennial weed, Elymus repens (target plant) and a forage grass, Festuca pratensis (non-target plant) and their arbuscular mycorrhizal fungal (AMF) root colonization in a field pot experiment. Non-target plants were sown in the following spring. Furthermore, we tested if glyphosate effects depend on tillage or soil properties modulated by long cultivation history of endophyte symbiotic grass (E+ grass). AMF root colonization, plant establishment and growth, glyphosate residues in plants, and soil chemistry were measured. Glyphosate reduced the mycorrhizal colonization and growth of both target and non-target grasses. The magnitude of reduction depended on tillage and soil properties due to cultivation history of E+ grass. We detected glyphosate residues in weeds and crop plants in the growing season following the glyphosate treatment. Residues were higher in plants growing in no-till pots compared to conspecifics in tilled pots. These results demonstrate negative effects of glyphosate on non-target organisms in agricultural environments and grassland ecosystems.

 Conclusion

Our results demonstrate that the effects of chemical and mechanical agricultural practices on ecosystem services can exceed the year of application. Here we showed that glyphosate negatively affects plant beneficial AMF in target weeds and non-target forage grasses. These results call attention to more comprehensive understanding of the effects of glyphosate on other plant associated microbes and microbial communities in soils. For example, soil microbes are known to play an important role in glyphosate degradation (Ayansina and Oso, 2006), and thus changes in their frequencies and community composition may affect the amount of available glyphosate for crop plants and weeds in the soil. Beside biotic factors also the abiotic environment is important in determining actions of the glyphosate in the soil. Depending on the initial soil microbiota, glyphosate may differently modify the outcome of microbial composition for glyphosate degradation and for the weeds and crop plants. Specifically in non-target F. pratensis AMF colonization was decreased interactively with biotic factors including the symbiotic fungus of the previous field cover plant, and abiotic factors including cultivation techniques. The core of a matter in maintaining fertile soils for sustainable crop production is to provide favorable environment for the soil and plant beneficial microbes.

چکیده

سابقه: کورستین (Q) یکی از فراوان ترین فلاونوئیدهای موجود در منابع غذایی انسان است و با ظرفیت بالقوه ی بیماری های مرتبط با چاقی در ارتباط است. کورستین-3-0-B-O- گلوکورونید (Q3GA) متابولیت اصلی در گردش خون است، اما افکتورهای نهایی داخل سلولی برای فعالیت آن هنوز مشخص نیست.

فرضیه/هدف: شناسایی و اندازه گیری متابولیت های داخل سلولی در آدیپوسیت های هیپرتروفی شده ی انکوبه شده با Q یا Q3GA و ارتباط دادن آن ها با تولید درون سلولی گونه های رادیکال اکسیژن (ROS).

روش ها: فرکشن های سیتوپلاسمی به دست آمده و متابولیت های کورستین با استفاده از کروماتوگرافی مایع همراه با یک آشکارساز time-of-flight با یونیزاسیون الکترواسپری (HPLC-DAD-ESI-TOF) تعیین شدند. تولید درون سلولی ROS با استفاده از یک پروب فلورسنت حساس به ROS اندازه گیری شد.

نتایج: Q و Q3GA هر دو به وسيله آديپوسيت هاي هيپرتروفي شده جذب شدند و تاحدی به ترتیب به Q3GA و Q متابوليزه شدند، اما جذب Q بيشتر (92/1- 03/0 میکروگرم/میکروگرم پروتئين) و سريع تر از Q3GA (12/0-0015/0 میکروگرم/میکروگرم پروتئين)، که منجر به افزایش غلظت درون سلولی آگلیکون شد. کاهش درون سلولی ROS با وجود فراوان ترین متابولیت یعنی کورستین ارتباط دارد.

نتيجه گيري: Q و Q3GA به خوبی به آديپوسيت هاي هيپرتروفي شده جذب شده و تا حدودي به ترتیب به Q3GA و Q متابوليزه شدند. کاهش درون سلولی ROS در یک مدل آدیپوسیت هیپرتروفی شده ی تیمار شده با Q یا Q3GA با فراوان ترین متابولیت درون سلولی ارتباط دارد. هر دو ترکیب ممکن است بتوانند به سایر اهداف درون سلولی رسیده و به فعالیت زیستی آنها کمک کنند.

نتیجه گیری

در نتیجه، Q و Q3GA هر دو با آدیپوسیت های هیپرتروفی شده جذب شده و به ترتیب با فعالیت های گلوکورونوترنسفراز و گلوکورونیداز به Q3GA و Q متابولیزه شدند. جذب Q نسبت به Q3GA کارآمدتر و سریعتر بوده و منجر به افزایش غلظت داخل سلولی آگیلکون شد. هر دو متابولیت Q و Q3GA مسئول کاهش درون سلولی ROS در آدیپوسیت های هایپرتروفی شده بودند. این نتایج برای نخستین بار حاکی از این است که هر دو متابولیت ممکن است ظرفیت پراکنش قوی رادیکال را داشته باشند که در آدیپوسیت های هیپرتروفی شده-ی تیمار شده با عصاره پلی فنولی H. sabdariffa مشاهده شده است (Herranz-Lopez و همکاران، 2012) و همچنین ممکن است به سایر اهداف داخل سلولی که به فعاليت زيستي آن ها کمک مي کنند نیز برسند.

Abstract

Background Quercetin (Q) is one of the most abundant flavonoids in human dietary sources and has been related to the capacity to ameliorate obesity-related pathologies. Quercetin-3-O-β-d-glucuronide (Q3GA) is supposed to be the main metabolite in blood circulation, but the intracellular final effectors for its activity are still unknown. Hypothesis/purpose To identify and quantitate the intracellular metabolites in hypertrophied adipocytes incubated with Q or Q3GA and to correlate them with the intracellular generation of oxygen radical species (ROS). Methods Cytoplasmic fractions were obtained and quercetin metabolites were determined by liquid chromatography coupled to a time-of-flight mass detector with electrospray ionization (HPLC-DAD-ESI-TOF). Intracellular ROS generation was measured by a ROS-sensitive fluorescent probe. Results Both Q and Q3GA were absorbed by hypertrophied adipocytes and metabolized to some extent to Q3GA and Q, respectively, but Q absorption was more efficient (1.92 ± 0.03 µg/µg protein) and faster than that of Q3GA (0.12 ± 0.0015 µg/µg protein), leading to a higher intracellular concentration of the aglycone. Intracellular decrease of ROS correlated with the presence of the most abundant quercetin metabolite. Conclusion Q and Q3GA are efficiently absorbed by hypertrophied adipocytes and metabolized to some extent to Q3GA and Q, respectively. The intracellular decrease of ROS in a hypertrophied adipocyte model treated with Q or Q3GA is correlated with the most abundant intracellular metabolite for the first time. Both compounds might be able to reach other intracellular targets, thus contributing to their bioactivity.

Conclusions

In conclusion, both Q and Q3GA were absorbed by hypertrophied adipocytes and metabolized to Q3GA and Q by glucuronosyltransferase and glucuronidase activity, respectively. Absorption was more efficient and faster for Q than Q3GA, leading to a higher intracellular concentration of the aglycone. Both metabolites Q and Q3GA were responsible for an intracellular decrease of ROS in hypertrophied adipocytes. These results support for the first time that both metabolites may account for the strong radical scavenging capacity observed in hypertrophied adipocytes treated with the polyphenolic extract of H. sabdariffa (Herranz-Lopez et al., 2012) and might also reach other intracellular targets that contribute to their bioactivity.

Abstract

Different protein sources from plant and animal origin viz., soybean flour (SF-AD) and soaked soybean (SS-AD), bee pollen (BP-AD), honey bee drone larvae (DL-AD) and frozen aphids (AF-AD) were assessed as an artificial diet of C. septempunctata in comparison to their natural diet live aphids. Highest growth index was obtained with SS-AD (4.05) diet almost similar to control (4.0), followed by SF-AD, BP-AD, DL-AD and AF-AD as 3.8, 2.4, 1.98 and 1.94 respectively. Similarly, data showed that success index of the adults fed on an artificial diet containing soybean (SS-AD = 1.03, SF-AD = 0.98) was almost similar to control (1.0), whereas the diets containing bee pollen, drone larvae, and frozen aphids was lower than control as 0.83, 0.84 and 0.78, respectively. Protein concentration was highest in live aphid (5.9 μg/ml) diet followed by SS-AD (5.6), SF-AD (5.2), BP-AD (3.4), AF-AD (2.2) and DL AD (1.3). The results indicated that soybean based diets were more or equivalently suitable to beetles for their growth and development likewise their natural prey and the efficiency of artificial diets was related to its protein concentration.

1 Introduction

Predatory beetles (Order Coleoptera) have a long history of importance as natural enemies of various phytophagous insect and mite pests. Many coleopteran predators, especially those in the family Coccinellidae (representing the lady beetles, ladybird beetles or ladybugs) have been used with moderate success in managing pest populations throughout the world (Hodek and Honek, 1996; Obrycki and Kring, 1998; Hodek and Evans, 2012). Predatory insects are important not only because they are major biological control agents in IPM program but also because they form important material for behavioral, bioecological and screening bioassay studies. Consequently, there is an ever increasing demand for large supplies of these insects in research, agriculture, and industry requires large-scale production of predatory insects. Since their natural food, the host/prey species, is not always available, it becomes necessary to develop alternative feeding media on which the insects can be reared and multiplied. Today, one challenge for greater use of coccinellids is to create cost-effective techniques to rear and stockpile (store) them. Many of the current rearing methods continue to depend on a tri-trophic system of rearing: the host plant, natural prey (herbivorous pest) and predator. This system is labor intensive and not cost effective. Hence there is a need to develop a targeted and specific mass rearing methodology for these very important predators using artificial diets. Use of artificial diets represents a step towards cost-effective rearing of beneficial insects (Cohen, 2004). Developing artificial diet is difficult but once optimized, usually are simple to prepare and easy to handle (Cohen and Smith, 1998). Success or failure of an artificial diet depends on various criteria of considerable effects on different life parameters of coleopteran insect which includes: development time, larval survival, adult emergence, fecundity, and adult life span (Riddick and Chen, 2014; Sun et al., 2017). Predators need a protein rich diet for their growth. Lipid can improve the yield and fecundity of adults of several entomophagous insects (Grenier, 2012). Utilization of an artificial diet, rather than factitious prey/foods or natural prey, would be a further progression in realizing the costeffective mass production of lady beetles (Riddick, 2009). Purified proteins such as soybean, casein from milk, gluten from wheat and albumin from eggs have been used in artificial diets to meet the protein requirement of insects (Sarwar and Saqib, 2010). Jokar and Zarabi (2014) suggested that the semi-artificial diet was the most accurate diet for mass rearing of a predator. The seven-spotted ladybird beetle, Coccinella septempunctata L. (Coleoptera: Coccinellidae) was introduced into North America from Europe. C. septempunctata is predaceous in its feeding habits. Proteins are therefore a major component of its food. Naturally, they fed upon aphids, thrips, spider mites and various soft-bodied insects and their eggs. In the scarcity of insects, they may also survive on pollen of different flowering plants. To rear C. septempunctata on a large scale, it is important to incorporate a rich protein source in the artificial diet. Both plants and animals and their products can meet this requirement. In the present study three diverse types of protein sources viz., soybean (flour and soaked seeds), bee pollen, drone larvae and frozen aphids were assessed for their efficiency in artificial diet for mass rearing of C. septempunctata in comparison to their natural diet live aphids.

Material and methods

Plant and insect material

A culture of radish was established in pots (3 gal) in glass house outside Zoology Department, Panjab University, Chandigarh. These plants were used to maintain the culture of aphids Myzus persicae Sulz. Aphids were used to feed C. septempunctata as a natural diet. The adults of C. septempunctata were collected from radish fields, Village – Chappar-chiri, Punjab, India. The adults were kept at a 23–25° temperature and 16:8 hour photoperiod and 50–70% relative humidity. The beetles were reared in a plastic container (470 × 350 × 170 mm) bottom lined with filter paper and top covered with a muslin cloth. Thin apple slices, water, and honey were provided to them inside the container. A plant twig and scrambled paper were also placed inside the box, which provided these beetles extra surface to roam around. The container was observed daily for eggs. The box was changed after two days and replaced with new one. The collected eggs were kept in a small petridish lined with filter paper. These eggs were kept inside incubator till hatching. The newly emerged larvae were fed on aphids for one day and taken for further use.

Protein source: frozen aphids and drone larvae (animal origin) M.

persicae were collected from greenhouse plants and frozen in vials. Drone larvae of Apis mellifera L. obtained from honey bee colonies maintained by the Department of Zoology, Panjab University, Chandigarh, India. Soybean (Plant origin): soybean is one of the richest protein sources of plant origin. Soybean protein was in two different forms as soybean flour and seeds, soaked for 24 h before making the artificial diet. Bee pollen (Plant origin modified by an insect): as collected from A. mellifera colonies, pollen was of mixed type. Pollen was collected from colonies of honey bee by installing pollen trap at the entrance of langstroth bee hive, which were placed in the fields of village Zirakpur, Patiala, India.

Artificial diets

Artificial diets were prepared by following methods described by Tiwari and Bhattacharya (1987). The common components of diet per 100 g of diet were honey, yeast, agar-agar, sorbic acid, methylparaben, sodium ascorbate, 10% formaldehyde, distilled water, and multi-vitamins. Diet 1 contained soybean flour as the test material in addition to components mentioned above. For diets 2, 3, and 4: soaked soybean seeds, bee pollen, and frozen aphids respectively, were used as protein source. Live aphid diet constituted the control. The detailed composition of the four diets is shown in Table 1.
