چکیده
هدف: ارزشیابی شیوع Staphylococcus aureus مقاوم چنددارویی (S.aureus) در محصولات لبنی

روش ها: جداسازی و شناسایی S.aureus در 3 محصولات غذایی لبنی انجام شد. جداسازی ها برای استعداد در آسیب پذیری برای 5 داروی ضدمیکروبی رایج مختلف آزمایش شد.

نتایج: برای 50 نمونه آزمایش شده، 5 (10 درصد) با S.aureus در معرض آلودکی قرار گرفت. متعاقباً 5 جداسازی تحت الگوی مقاومت ضدمیکروبی با استفاده از 5 قرص آنتی بیوتیک قرار گرفت (متاسیلین، وانکومیسین, کانامیسین، کلروفنیکول و تتراسیلین). 29 نمونه مقاومت متی سیلین و وانکومیسین را نشان داد. 18 نمونه پاسخ میانه ای را برای تتراسیلین نشان داد. دیگر نمونه ها برای تمام انتی بیوتیک های آزمایش شده مورد تردید بودند.

نتیجه گیری: نتایج داده های اولیه برای منبع آلودگی غذایی را فراهم می نماید که می تواند به عنوان وسایلی برای انتقال Staphylococcus مقاومت-ضدمیکروب عمل نماید. بنابراین ما را برای توسعه راهبردهای پیشگیرانه برای اجتناب از حضور آسیب های جدید برای S.aureus مقاوم توانا می سازد.

1- مقدمه
Staphylococcus aureus) S. aureus) یک پاتوژن در انسان ها و حیوانات است که مسبب طیف وسیعی از بیماری ها در گستره نقایص پوستی مینور تا بیماری های تهدید کننده عمر انسان، از قبیل ذات الریه و مننژیت است. همچنین به عنوان یکی از مهمترین عوامل مسمویت غذایی در دنیا شناخته شده است. این باور وجود دارد که تمام آسیب های باکتریایی با گستره وسیعی از عوامل ضدمیکروبی و موثر قابل درمان هستند. هرچند، مقاومت برای انتی بیوتیک های چندگانه در بین Staphylococcus aureus(S. aureus) اخباری جدید برای محققان و اقدام کنندگان در مورد سلامتی بشر است. گزارش شده که کمی بعد از عرضه پنی سیلین در دهه 1940، مقاومت گسترش یافته در مقابل Staphylococcus aureus(S. aureus) توسط مقامومت به متی سیلین و اخیراً گلی سوپپتاید مثلاً وانکومیسین دنبال شد.
Staphylococcus aureus(S. aureus) می تواند به انسان ها از طریق شیر الوده شده و محصولات شیر انتقال یابد. S. aureus در پوسته و مخاط باقیمانده های تولیدات حیوانی حاضر می شود که شامل جانوران پستاندارد می شود و اغلب به غده های پستان بالینی و بالینی فرعی منجر به آلودکی محصولات لبنی می شود. این باکتری به صورت پاتوژن عمده ایجاد شده رایج در غده پستان گاو در نظر گرفته می شود.
هدف از این مطالعه جداسازی S. aureus از محصولات لبنی است که شامل شیر تازه گاو، شیر پاستوریزه شده، ماست و پنیر برای مشخص سازی S. aureus با استفاده از روش های رایج مانند کشش Gram ازمون کواگالاز و آزمون کاتالاز و همچنین تعیین شیوع مقاومت ضد میکروبی در بین محصولات لبنی جدا شده می شود.

Abstract

Objective To evaluate the prevalence of multidrug resistant Staphylococcus aureus (S. aureus) in dairy products.

Methods Isolation and identification of S. aureus were performed in 3 dairy-based food products. The isolates were tested for their susceptibility to 5 different common antimicrobial drugs.

Results Of 50 samples examined, 5 (10%) were contaminated with S. aureus. Subsequently, the 5 isolates were subjected to antimicrobial resistance pattern using five antibiotic discs (methicillin, vancomycin, kanamycin, chloramphenicol and tetracycline). Sample 29 showed resistance to methicillin and vancomycin. Sample 18 showed intermediate response to tetracycline. The other samples were susceptible to all the antibiotics tested.

Conclusions The results provide preliminary data on sources of food contamination which may act as vehicles for the transmission of antimicrobial-resistant Staphylococcus. Therefore, it enables us to develop preventive strategies to avoid the emergence of new strains of resistant S. aureus.

1- Introduction

Staphylococcus aureus(S. aureus) is a versatile pathogen in humans and animals which is responsible for a diverse spectrum of diseases ranging from minor skin infections to life threatening diseases, such as pneumonia and meningitis[10]. It is also known as one of the most important agents of food poisoning globally[7]. It has been believed that all bacterial infections were treatable with a vast array of effective and antimicrobial agents. However, the emergence of resistance to multiple antibiotics among S. aureus has created breaking news for health practitioners and researchers[15]. It has been reported that shortly after the introduction of penicillin in 1940s, resistance developed in S. aureus, followed by resistance to methicillin and more recently to glycopeptides, e.g. ,vancomycin[11].

S. aureus can be transmitted to humans through contaminated and untreated milk and milk products[14]. S. aureus presents on the skin and mucosae of food producing animal reservoirs that include ruminants and it is frequently associated with sub-clinical or clinical mastitis leading to the contamination of dairy products[8]. This bacterium is considered as the most commonly occurring major pathogen of cow’s mammary gland[5].

The objective of this study was to isolate S. aureus from dairy products which include fresh cow milk, pasteurized milk, goat milk, yogurt and cheese, to characterize S. aureus using conventional methods like Gram staining, coagulase test and catalase test and also to determine the prevalence of antimicrobial resistance among S. aureus isolated from dairy products..
