چکیده

قرارگیری در معرض حشره‌کش ممکن است آثار میان‌نسلی و چندنسلی را در جمعیت‌ها ایجاد نماید، اما مکانیسم‌های مولکولی این تغییرات تا حد زیادی ناشناخته باقی‌مانده است. اکثر مطالعات بر روی مکانیسم‌های میان‌نسلی یا چند‌نسلی متمرکز شده‌اند اما از جنبه‌های مقایسه‌ای آنها غافل مانده‌اند. این مطالعه بررسی کرد که آیا حشره‌کش پیرتیروئید اتوفن‌پراکس (فرمولاسیون EC 30 تربون®) آثار میان‌نسلی و یا چند‌نسلی بر روی بقا و تولید مثل فولسومیا کاندیدا(کولمبولا) دارد یا خیر. فعال سازی ژن‌های وابسته به تنش برای کشف اینکه اتوفن‌پراکس بیان ژن‌های وابسته به تولید مثل را در تیمارهای میان و چند‌نسلی تغییر می‌دهد یا خیر، بررسی گردید. یک مطالعه‌ی آزمایشگاهی برای سه نسل با غلظت‌های حشره‌کش در خاک لوفا2.2 انجام گرفت. در تیمار چند نسلی، تنها نسل والدین (P) مورد بررسی قرار گرفت، اما برای نسل‌های بعدی چنین نبود. در تیمار چندنسلی، هر سه نسل در یک روش مشابه در معرض حشره‌کش قرار گرفتند. قرارگیری در معرض حشره‌کش موجب کاهش آثار تولیدمثل در طول نسل‌ها می‌شود و پیشنهاد می‌شود که فولسومیا کاندیدا قادر به سازگاری با افزایش سطح غلظت اتوفن‌پراکس در طولانی مدت در طی چندین نسل است. در تیمار میان‌نسلی، بیان پروتئین شوک حرارتی 70 در بالادست و سیتوکروم اکسیداز 1v4N6 در پایین دست در دوز وابسته به نسل F2 تنظیم گردید. این یافته‌ها احتمال وراثت اپی‌ژنتیکی آثار حشره‌کش بر والدین را بالا می‌برد. بعلاوه، بیان سیتوکروم اکسیداز 1v4N6 برخلاف تیمارهای میان و چند‌نسلی تنظیم شد. نتایج ما توجهات را به سمت اختلاف پاسخ‌ها در سطح مولکولی فلوسمیا کاندیدا به قرارگیری در معرض اتوفن‌پراکس میان و چند‌نسلی جلب می‌نماید.

5- نتیجه‌گیری

حشره‌کش اتوفن‌پراکس، در فرمولاسیون تربون ® 30EC، نشان داد که در دو نسل میان نسلی و چندنسلی بر روی فلوسومیا کاندیدا اثر داشته است. در تیمار میان نسلی، اثرات روی زنده‌ها و تولیدمثلی در نسل اول مشاهده شد، اما این یافته ممکن است ناشی از قرارگیری مستقیم نابالغ‌های نسل اول در معرض حشره‌کش باشد. اثرات تراریخته‌ی ظاهری اتوفن‌پراکس در رونویسی پروتئین شوک گرمایی 70 و سیتوکروم اکسیداز 1v4N6 در نسل دوم بدست آمد. بیان پروتئین شوک گرمایی70 در بالادست و سیتوکروم اکسیداز 1v4N6در پایین دست به صورت وابسته به دوز و بدون اثر مستقیم آفت‌کش تنظیم شدد. مطالعه‌ی بیشتر مکانیسم‌های مولکولی در پس تغییرات بیان سیتوکروم اکسیداز 1v4N6و پروتئین شوک گرمایی70 نیاز است. اثرات چندنسلی به عنوان اثر کاهشی روی تولیدمثل (تعداد نابالغ‌ها) در نسل اول و دوم بیان شده است. پاسخ‌های سلولی به اتوفن‌پراکس در تیمارهای میان نسلی و چندنسلی ظاهر شد و حساسیت بالایی را نشان داد. مقدار RNAی بدست آمده در تمام تیمارها به طور وابسته به دوز کاهش یافت(به جز در نسل میان نسلی2). که حاکی از ضعف سلامتی کولمبولانس‌های در معرض اتوفن‌پراکس است. الگوهای ترانسکریپتومی ژن‌های تنش می‌تواند حاکی از تنش اتوفن‌پراکس در جمعیت باشد. ژن پروتئین شوک گرمایی70 نشان دهنده‌ی تنش عمومی است اما ژن سیتوکروم اکسیداز 1v4N6در ارتباط با سم‌زدایی زنوبیوتیک‌های دارای پیوند استری است. ژن‌های فعال شده در تیمار میان نسلی و کاهش مهار تولیدمثل در تیمار چندنسلی مشابه به مرحله‌ی اول مقاومت یا سازگاری است.

Abstract

Insecticide exposure may cause both transgenerational and multigenerational effects on populations, but the molecular mechanisms of these changes remain largely unclear. Many studies have focused on either transgenerational or multigenerational mechanisms but did neglect the comparative aspects. This study assessed whether the pyrethroid insecticide etofenprox (formulation Trebon® 30 EC) shows transgenerational and/or multigenerational effects on the survival and reproduction of Folsomia candida (Collembola). The activation of stress-related genes was studied to detect whether etofenprox modifies the expression of reproduction-associated genes in trans- and multigenerational treatments. A laboratory study was carried out for three generations with five insecticide concentrations in LUFA 2.2 soil. In the transgenerational treatment, only the parent generation (P) was exposed, but the subsequent generations were not. In the multigenerational treatment, all three generations were exposed to the insecticide in the same manner. Multigenerational exposure resulted in reduced reproduction effects over generations, suggesting that F. candida is capable of acclimating to enhanced concentration levels of etofenprox during prolonged exposure over multiple generations. In the transgenerational treatment, the heat shock protein 70 was up-regulated and cytochrome oxidase 6N4v1 expression down-regulated in a dose-dependent manner in the F2 generation. This finding raises the possibility of the epigenetic inheritance of insecticide impacts on parents. Furthermore, CYP6N4v1 expression was oppositely regulated in the trans- and multigenerational treatments. Our results draw attention to the differences in molecular level responses of F. candida to trans- and multigenerational etofenprox exposure.

5- Conclusions

The insecticide etofenprox, in the formulation Trebon® 30 EC, showed both transgenerational and multigenerational effects on F. candida. In the transgenerational treatment, effects on survival and reproduction were observed in the F1 generation, but this finding may be due to direct exposure of the F1 juveniles to the insecticide. Apparent transgenerational effects of etofenprox on the transcription of HSP70 and CYP6N4v1 were found in the F2 generation. The HSP70 expression was upregulated and the CYP6N4v1 down-regulated in a dose-related manner without direct pesticide exposure. Further study of the molecular mechanisms behind the observed HSP70 and CYP6N4v1 expression changes is required. Multigenerational effects are expressed as a reduced effect on reproduction (juvenile numbers) at F1 and F2 generations. Cellular responses to etofenprox appeared both in transand multigenerational treatments, and showed high sensitivity. The obtained amount of RNA was decreased in all treatments in a doserelated manner (except in TF2 generation), suggesting poor health of the etofenprox-exposed collembolans. The stress-connected genes reacted with a dose-related increase to etofenprox exposure, which suggests that the population could be more resistant to further insecticide exposure or other environmental stress factors. The transcriptomic patterns of the stress genes could indicate an etofenprox stress of the population. The HSP70 gene indicates general stress, but the CYP6N4v1 gene is connected with the detoxification of ester bond-containing xenobiotics. The activated genes in the transgenerational treatment and the reduced inhibition of reproduction in the multigenerational treatment both resemble an early stage of resistance or acclimation.

Abstract

Different protein sources from plant and animal origin viz., soybean flour (SF-AD) and soaked soybean (SS-AD), bee pollen (BP-AD), honey bee drone larvae (DL-AD) and frozen aphids (AF-AD) were assessed as an artificial diet of C. septempunctata in comparison to their natural diet live aphids. Highest growth index was obtained with SS-AD (4.05) diet almost similar to control (4.0), followed by SF-AD, BP-AD, DL-AD and AF-AD as 3.8, 2.4, 1.98 and 1.94 respectively. Similarly, data showed that success index of the adults fed on an artificial diet containing soybean (SS-AD = 1.03, SF-AD = 0.98) was almost similar to control (1.0), whereas the diets containing bee pollen, drone larvae, and frozen aphids was lower than control as 0.83, 0.84 and 0.78, respectively. Protein concentration was highest in live aphid (5.9 μg/ml) diet followed by SS-AD (5.6), SF-AD (5.2), BP-AD (3.4), AF-AD (2.2) and DL AD (1.3). The results indicated that soybean based diets were more or equivalently suitable to beetles for their growth and development likewise their natural prey and the efficiency of artificial diets was related to its protein concentration.

1 Introduction

Predatory beetles (Order Coleoptera) have a long history of importance as natural enemies of various phytophagous insect and mite pests. Many coleopteran predators, especially those in the family Coccinellidae (representing the lady beetles, ladybird beetles or ladybugs) have been used with moderate success in managing pest populations throughout the world (Hodek and Honek, 1996; Obrycki and Kring, 1998; Hodek and Evans, 2012). Predatory insects are important not only because they are major biological control agents in IPM program but also because they form important material for behavioral, bioecological and screening bioassay studies. Consequently, there is an ever increasing demand for large supplies of these insects in research, agriculture, and industry requires large-scale production of predatory insects. Since their natural food, the host/prey species, is not always available, it becomes necessary to develop alternative feeding media on which the insects can be reared and multiplied. Today, one challenge for greater use of coccinellids is to create cost-effective techniques to rear and stockpile (store) them. Many of the current rearing methods continue to depend on a tri-trophic system of rearing: the host plant, natural prey (herbivorous pest) and predator. This system is labor intensive and not cost effective. Hence there is a need to develop a targeted and specific mass rearing methodology for these very important predators using artificial diets. Use of artificial diets represents a step towards cost-effective rearing of beneficial insects (Cohen, 2004). Developing artificial diet is difficult but once optimized, usually are simple to prepare and easy to handle (Cohen and Smith, 1998). Success or failure of an artificial diet depends on various criteria of considerable effects on different life parameters of coleopteran insect which includes: development time, larval survival, adult emergence, fecundity, and adult life span (Riddick and Chen, 2014; Sun et al., 2017). Predators need a protein rich diet for their growth. Lipid can improve the yield and fecundity of adults of several entomophagous insects (Grenier, 2012). Utilization of an artificial diet, rather than factitious prey/foods or natural prey, would be a further progression in realizing the costeffective mass production of lady beetles (Riddick, 2009). Purified proteins such as soybean, casein from milk, gluten from wheat and albumin from eggs have been used in artificial diets to meet the protein requirement of insects (Sarwar and Saqib, 2010). Jokar and Zarabi (2014) suggested that the semi-artificial diet was the most accurate diet for mass rearing of a predator. The seven-spotted ladybird beetle, Coccinella septempunctata L. (Coleoptera: Coccinellidae) was introduced into North America from Europe. C. septempunctata is predaceous in its feeding habits. Proteins are therefore a major component of its food. Naturally, they fed upon aphids, thrips, spider mites and various soft-bodied insects and their eggs. In the scarcity of insects, they may also survive on pollen of different flowering plants. To rear C. septempunctata on a large scale, it is important to incorporate a rich protein source in the artificial diet. Both plants and animals and their products can meet this requirement. In the present study three diverse types of protein sources viz., soybean (flour and soaked seeds), bee pollen, drone larvae and frozen aphids were assessed for their efficiency in artificial diet for mass rearing of C. septempunctata in comparison to their natural diet live aphids.

Material and methods

Plant and insect material

A culture of radish was established in pots (3 gal) in glass house outside Zoology Department, Panjab University, Chandigarh. These plants were used to maintain the culture of aphids Myzus persicae Sulz. Aphids were used to feed C. septempunctata as a natural diet. The adults of C. septempunctata were collected from radish fields, Village – Chappar-chiri, Punjab, India. The adults were kept at a 23–25° temperature and 16:8 hour photoperiod and 50–70% relative humidity. The beetles were reared in a plastic container (470 × 350 × 170 mm) bottom lined with filter paper and top covered with a muslin cloth. Thin apple slices, water, and honey were provided to them inside the container. A plant twig and scrambled paper were also placed inside the box, which provided these beetles extra surface to roam around. The container was observed daily for eggs. The box was changed after two days and replaced with new one. The collected eggs were kept in a small petridish lined with filter paper. These eggs were kept inside incubator till hatching. The newly emerged larvae were fed on aphids for one day and taken for further use.

Protein source: frozen aphids and drone larvae (animal origin) M.

persicae were collected from greenhouse plants and frozen in vials. Drone larvae of Apis mellifera L. obtained from honey bee colonies maintained by the Department of Zoology, Panjab University, Chandigarh, India. Soybean (Plant origin): soybean is one of the richest protein sources of plant origin. Soybean protein was in two different forms as soybean flour and seeds, soaked for 24 h before making the artificial diet. Bee pollen (Plant origin modified by an insect): as collected from A. mellifera colonies, pollen was of mixed type. Pollen was collected from colonies of honey bee by installing pollen trap at the entrance of langstroth bee hive, which were placed in the fields of village Zirakpur, Patiala, India.

Artificial diets

Artificial diets were prepared by following methods described by Tiwari and Bhattacharya (1987). The common components of diet per 100 g of diet were honey, yeast, agar-agar, sorbic acid, methylparaben, sodium ascorbate, 10% formaldehyde, distilled water, and multi-vitamins. Diet 1 contained soybean flour as the test material in addition to components mentioned above. For diets 2, 3, and 4: soaked soybean seeds, bee pollen, and frozen aphids respectively, were used as protein source. Live aphid diet constituted the control. The detailed composition of the four diets is shown in Table 1.

نتایج و بحث
نتایج حاصل از این تحلیل ها نشان داد که تنها پاتوژنی که در حال حاضر در تمام نمونه های آنالیز شده ی زنبور عسل وجود داشت، Noserna cera (میکروسپوریدی) بود (خلاصه شده در جدول 1). بررسی ها نشان داد که به دلیل آلودگی شدید سلول های اپیتلیومی، معده متورم و شکننده شده و ویژگی هایی مشابه با ویژگی های آلودگی N. ceranae را نشان داد (Higes et at., 2007; 2008b). شواهدی از تخریب سلول های اپیتلیومی و فساد گسترده وجود داشت (شکل های 3 و 4). همان طور که قبلا توضیح داده شد (Higes et al., 2008a)، سلول-های به شدت آلوده ممکن است بمیرند یا در حال مرگ باشند، که در نهایت موجب مرگ زودهنگام زنبورهای عسل کارگر آلوده به دلیل گرسنگی خواهند شد (Liu, 1984). این قضیه ممکن است منجر به عدم بازگشت زنبورهای به شدت آلوده به کندو شده (ترجیحا زنبورهای جستجوگر) و در نهایت، به دلیل عدم جبران از بین رفتن زنبورهای جستجوگر توسط ملکه، کاهش جمعیت در کلنی مشهود است، کاهش جمعیت آشکار شده و منجر به مرگ می شود (Higes et al., 2008a).

Abstract

Honeybee colony collapse is a sanitary and ecological worldwide problem. The features of this syndrome are an unexplained disappearance of adult bees, a lack of brood attention, reduced colony strength, and heavy winter mortality without any previous evident pathological disturbances. To date there has not been a consensus about its origins. This report describes the clinical features of two professional bee-keepers affecting by this syndrome. Anamnesis, clinical examination and analyses support that the depopulation in both cases was due to the infection by Nosema ceranae (Microsporidia), an emerging pathogen of Apis mellifera. No other significant pathogens or pesticides (neonicotinoids) were detected and the bees had not been foraging in corn or sunflower crops. The treatment with fumagillin avoided the loss of surviving weak colonies. This is the first case report of honeybee colony collapse due to N. ceranae in professional apiaries in field conditions reported worldwide.

Results and discussion

The results of these analyses indicated that the only pathogen present in all the honeybee samples analysed was Nosema ceranae (Microsporidia) (as summarized in Table 1). The gross overview of the ventriculus showed them to be oedematous and friable, due to a strong epithelial cell infection, and with similar features to those described previously in association with N. ceranae infection (Higes et al., 2007; 2008b). There was evidence of epithelial cell degeneration and extensive lysis (Figs 3 and 4). As described previously (Higes et al., 2008a), the heavily infected cells may be either dead or dying, which will eventually lead to the early death of the infected worker bees due to starvation (Liu, 1984). This leads to the result that some heavily infected honeybees (foragers preferably) do not return to the hive and finally the depopulation of colonies is evident when the queen cannot compensate for the loss of foragers, depopulation becomes evident and death is forthcoming (Higes et al., 2008a).

Summary

Nosema ceranae is an emergent and potentially virulent pathogen of the honey bee (Apis mellifera) that has spread across the world in the last 10 or so years. Its precise origin and timing of spread are currently unclear because of a lack of appropriate genetic markers and inadequate sampling in putative Asian source populations. Though it has been dismissed as a cause of CCD in the USA based on correlational analyses of snapshot sampling of diseased hives, observations of naturally infected colonies suggest that it leads to colony collapse in Spain. Experiments are sorely needed to investigate its impact on individuals and colonies, and to pin down a causal relationship between N. ceranae and colony collapse. Whether N. ceranae is displacing N. apis is uncertain. For temperate zone apiculturalists, global climate change may mean that N. ceranae presents more of a challenge than has hitherto been considered the case.

Outlook

Nosema ceranae is clearly a novel, emergent pathogen of A. mellifera with potentially very serious effects on the individual and colony. Whether it causes CCD, alone or in combination with other synergistic factors such as habitat loss and lack of floral resources, is still an open question. Under a scenario of global climate change, N. ceranae may exert an increasing impact on world beekeeping with A. mellifera, analogous to the impact of emergent fungal pathogens tied to global warming on amphibian populations (Pounds et al., 2006). Research is sorely needed (i) to develop genetic markers for N. ceranae to differentiate intraspecific strains and (ii) to experimentally demonstrate a causal link between N. ceranae and CCD. That N. ceranae can be controlled by the fungicide fumagillin is of some reassurance for apiculture with A. mellifera (Martín-Hernandéz et al., 2007; Higes et al., 2008; 2009b; Williams et al., 2008b).

Abstract

Honey bee samples collected between 1995 and 2007 from 12 states were examined for the presence of Nosema infections. Our results showed that Nosema ceranae is a wide-spread infection of the European honey bee, Apis mellifera in the United States. The discovery of N. ceranae in bees collected a decade ago indicates that N. ceranae was transferred from its original host, Apis cerana to A. mellifera earlier than previously recognized. The spread of N. ceranae infection in A. mellifera warrants further epidemiological studies to identify conditions that resulted in such a widespread infection.

Nosema disease is one of the most prevalent adult honey bee diseases (Bailey, 1981; Matheson, 1993) and is caused by two described species of microsporidia, Nosema apis and Nosema ceranae. Transmission of Nosema in honey bee colonies is mainly via the fecal-oral route in which pathogens are spread by transferring feces of diseased hosts to uninfected hosts via ingestion. Adult bees ingest Nosema spores when they are eating contaminated food and when they are cleaning up fecal material from infected bees. The spores geminate within the midgut and release polar tubes that transfer their sporoplasm into midgut epithelial cells where they generate more spores. Millions of new spores can be found inside of a bee’s midgut a few weeks after initial infection (Bailey and Ball, 1991) and the spores excreted with feces become new sources of the infection in the colonies. Although infected bees do not exhibit obvious external disease symptoms, infection of Nosema causes digestive disorders, shortened life spans of honey bees, decrease of population size of honey bee colonies (Hassanein, 1953; Rinderer and Sylvester, 1978; Malone et al., 1995), and reduction of honey production and crop products that rely on bees for pollination (Anderson and Giacon, 1992; Fries et al., 1984; Goodwin et al., 1990).

Abstract

Using quantitative PCR that amplified a prey-specific mtDNA 214 bp amplicon from the COI mitochondrial gene of the Colorado potato beetle, Leptinotarsa decemlineata (Say) (Coleoptera: Chrysomelidae), prey eggs of known age and number were fed to larvae of the generalist predator lady beetle Coleomegilla maculata (De Geer) (Coleoptera: Coccinellidae), to elucidate the effects of time and diet since consumption, number of prey eggs, and methods for sample fixation and preservation, on the quantity of target DNA detected . Signal was strongly attenuated directly after cessation of feeding, even when predators were immediately frozen at −20°C. However, the quantity of target detected was significantly related to the number of eggs consumed and the time elapsed time since eating. Decrease in detected prey DNA was consistent with a negative exponential model. The target DNA sequence disappeared from starved predators (quantitative half-life estimate of 59 min) more slowly than those fed potato aphids after consuming the target prey eggs (half-life estimate 16 min), whereas those fed C. maculata eggs as a chaser were intermediate in the rate at which they degraded the target prey DNA sequence.
