چکیده

اعتیاد به داروها مانند مواد مخدر نتیجه تعامل فاکتورهای محیطی و ژنتیکی است. نشان داده‌اند که فرزند افراد معتاد خطر بالاتری به اعتیاد دارویی دارند. با این وجود، سازوکارهایی مانند اثرات وراثتی (بین نسلی) داروها مشخص نیست. در این مطالعه اثرات مصرف مورفین در والدین روی اضطراب، ترجیح مورفین و بیان mRNA گیرنده‌های دوپامین توله موش‌های پسر  F1و  F2 بررسی کرده‌ایم. مورفین بصورت بلند مدت به موش‌های نر بالغ و موش‌های ویستار ماده داده شد و 14 روز قبل از جفت‌گیری مصرف آن قطع شد. ترجیح مورفین و رفتار اضطرابی در موش‌های پسر به ترتیب با پارادایم انتخاب دو بطری و ماز صلیبی شکل مرتفع اندازه‌گیری شدند. برای اندازه‌گیری میزان بیان mRNA گیرنده‌های دوپامین در جسم مخطط، ﻧﻮﻛﻠﺌـﻮس اﻛـﻮﻣﺒﻨﺲ، قشر پیش فرونتال و هیپوکامپ حیوانات F1 از Real-time PCR استفاده شد. یافته‌ها نشان داد که موش‌های پسر F1 والدین مواجهه یافته با مورفین در مقایسه با پسرموش‌های والدینی که تحت درمان سالین قرار گرفته بودند ترجیح مورفین و رفتارهای اضطرابی بیشتری داشتند. در پسرموش‌های F1 والدین مورفینی، گیرنده‌های دوپامین D1 و D5 بطور قابل توجهی در قشر پیش فرونتال و ﻧﻮﻛﻠﺌـﻮس اﻛـﻮﻣﺒﻨﺲ افزایش یافت. گیرنده‌های D5 و D2 در هیپوکامپ کاهش یافت. گیرنده دوپامین D4 در جسم مخطط و هیپوکامپ افزایش و در قشر پیش فرونتال کاهش یافت. مواجهه مزمن موش‌های بزرگسال نر و ماده با مورفین قبل از لقاح منجر به بالاتر رفتن ترجیح مورفین و افزایش اضطراب در پسرموش‌های F1 شد اما این حالات در F2 مشاهده نشد. تغییرات بیان گیرنده دوپامین در سیستم پاداش شاید یک مکانیسم دخیل در بروز تغییرات پسر موش‌های F1 باشد. 

1. مقدمه 

اعتیاد به مواد مخدر مانند مورفین اختلالی مزمن است که بر مسیر مزوکورتیکولیمبیک دوپامینرژیک در مغز تاثیر می‌گذارد. در نهایت فعالسازی مسیر منجر به رهاسازی دوپامین در ﻧﻮﻛﻠﺌـﻮس اﻛـﻮﻣﺒﻨﺲ (NAc) (دی چیارا و همکاران، 1998) و تحریک گیرنده‌های دوپامین می‌شود. این گیرنده‌ها در دو دسته اصلی قرار می‌گیرند: گیرنده‌های D1 مانند (D1 و D5) و D2 مانند (D2، D3 و D4) که هر یک از  ژن متفاوتی تولید می‌شوند (سیبلی و همکاران 1993). فعالسازی گیرنده‌های دوپامینی D1- مانند D2- مانند به ترتیب باعث کاهش انگیزه یافتن پاداش و تسهیل خصوصیات تقویتی مواد مخدر اعتیاد آور می‌شوند (لی و همکاران، 2006).

5. نتیجه‌گیری

داده‌های حاصل از این مطالعه نشان داد که مواجهه مزمن والدین با مورفین در موش‌های بالغ قبل از لقاح منجر به ترجیح بالاتر مورفین و افزایش اضطراب در فرزندان پسر F1 می‌شود اما در F2 این چنین نیست. بیان این نکته حائز اهمیت است که مواجهه با مواد مخدر طی بارداری در حیوانات F1 وجود نداشت زیرا مصرف مواد مخدر در والدین دو هفته قبل از جفت‌گیری به صفر رسید. علاوه بر این، یافته‌های ما نشان داد که تغییرات در مشخصات بیان گیرنده‌های دوپامین در مسیر پاداش ممکن است یکی از سازوکارهای دخیل در تغییرات مشاهده شده در حیوانات F1 باشد که می‌تواند عامل ابتلاء به سوء استعمال مواد مخدر در فرزند حیوانات مواجهه یافته با مورفین باشد. 

Abstract

Addiction to drugs, including opioids is the result of an interplay between environmental and genetic factors. It has been shown that the progeny of addict people is at higher risk for drug addiction. However, the mechanisms of such trans-generational effects of drugs are not so clear. Here we have evaluated the effects of parental morphine consumption on anxiety, morphine preference, and mRNA expression of dopamine receptors in F1 and F2 male offspring. Morphine was chronically administered to adult male and female Wistar rats followed by 14-day abstinence before mating. Morphine preference and anxiety-like behavior in the offspring were measured by two-bottle-choice paradigm and elevated-plus maze, respectively. Real-time PCR was used to measure the mRNA expression level of dopamine receptors in the striatum, nucleus accumbens, prefrontal cortex, and hippocampus of F1 animals. The results indicated that F1 but not the F2 male progeny of morphine-exposed parents had a greater preference for morphine, and more anxiety-like behavior compared to the offspring of saline-treated parents. In F1 male progeny of morphine-treated parents, D1 and D5 dopamine receptors were significantly increased in the prefrontal cortex and nucleus accumbens. D5 and D2 receptors were decreased in the hippocampus. D4 dopamine receptor was augmented in striatum and hippocampus and decreased in the prefrontal cortex. Adulthood exposure to chronic morphine in male and female rats before conception leads to higher morphine preference and increased anxiety in F1 but not F2 male progeny. Alterations of dopamine receptor expression in the reward system may be one mechanism responsible for observed changes in F1 offspring.

1. Introduction

Addiction to opioids such as morphine is a chronic disorder that affects the mesocorticolimbic dopaminergic pathway in the brain. Activation of the pathway ultimately leads to dopamine release in the nucleus accumbens (NAc) (Di Chiara et al., 1998) and stimulation of dopamine receptors. These receptors are considered as two main categories: D1-like (D1 and D5) and D2-like (D2, D3, and D4) receptors, each of which is generated from a different gene (Sibley et al., 1993). Activation of D1-like and D2-like dopamine receptors decreases motivation to seek reward or facilitate the reinforcing properties of addictive drugs, respectively (Li et al., 2006).

5. Conclusion

The data of the present study revealed that chronic parental morphine exposure of adult rats before gestation results in higher morphine preference and increased anxiety in their F1 but not F2 male offspring. It is important to highlight that there was no gestational exposure to opioids in the F1 animals because parents were remained drug-free for two weeks prior to mating. Furthermore, our results showed that alterations in the expression profile of dopamine receptors in the reward pathway might be one of the mechanisms involved in observed changes in F1 animals, which could account for susceptibility to opioid abuse in the litter of morphine-exposed animals.

چکیده

هدف: افسردگی کموربید در میان بیماران مبتلا به صرع رایج می باشد.به هر حال،افسردگی مرتبط با صرع همیشه  غیر معمول است و به طور کامل توسط متخصصان اعصاب شناسایی نشده است.هدف این تحقیق، مقایسه تغییرات رفتاری و پاتولوژیکی بین مدل صرع در موش های دارای لیتیوم کلرید-پیلوکارپین مزمن ( مدل LiCl-فیلوکارپین) ومدل افسردگی  مزمن غیرقابل پیش بینی موش (مدلCUMS) است و تلاش می کند تااختلافات بین افسردگی اولیه و افسردگی مرتبط با صرع را بیاید.

روش ها: مدل CUMS و مدل LiCl-فیلوکارپین به ترتیب و همزمان ارائه شدند.حملات همزمان با کنترل و نظارت ویدئویی ضبط شدند.آزمون شنای اجباری FST و آزمون مصرف مواد شیرین SCT برای بررسی رفتارهای افسردگی انجام شدند.زمان بی تحرکیIMT و زمان بالارفتنCMT در FST – اولویت مواد شیرین موش ها SPR در SCT و وزن به دست آمده برای موش WGR برای نشان دادن شدت رفتارهای افسردگی مورد استفاده قرار می گیرند.چرک کردن ایمنوفلورسانس نیز برای اندازه گیری و سنجش پروتئین های هسته ای NeuN- پروتئین اسیدی گلیایی فیبریلارین GFAPو CD11b در هیپوکامپ در گروه کنترل – مدل مدل CUMS و مدل LiCl-فیلوکارپین انجام شد.

نتایج :IMT طولانی تر در هر دو مدل مدل CUMS (p b 0.01) و مدل LiCl-فیلوکارپین (p b 0.05)  در مقایسه با گروه کنترل مشاهده می شود.اما کاهش WGR فقط در مدل CUMS مشاهده می شود.درصد موش های دارای CMT بیش از 100s می باشد که برای گروه LiCl-فیلوکارپین بیشتر از مدل (p b 0.05). CUMS است. افزایش CMT در مدل LiCl-فیلوکارپین در نمونه های دارای افسردگی خفیف (EMD, I M T ≤ 100 s) در مقایسه یا گروه کنترل مشاهده شد.افت ذهنی در هر دو مدل CUMS و مدل LiCl-فیلوکارپین در مقایسه با گروه کنترل مشاهده شد(.(p b 0.05به هر حال،تعداد سلول های GFAP و CD11b در مدل LiCl-فیلوکارپین بیشتر از مدل CUMS و کنترل است(p b 0.05)..

نتیجه گیری : اختلافاتی بین تغییرات رفتاری و پاتولوژیکی در دو مدل صرع در موش های دارای لیتیوم کلرید-پیلوکارپین مزمن ( مدل LiCl-فیلوکارپین) ومدل افسردگی  مزمن غیرقابل پیش بینی موش (مدلCUMS)  وجود دارد و تنها استثنا مربوط به بعضی از ویژگی های عمومی است.فعالسازی گلیوسیز و میکروگلیا در افسردگی مرتبط با بیماری صرع بیشتر از افسردگی اولیه مشاهده می شود.

5.نتیجه گیری ها

در این تحقیق، از مدل های CUMS و مدل LiCl-فیلوکارپین برای نشان دادن مدل افسردگی مرتبط با صرع و افسردگی اولیه استفاده کردیم و به این مطلب پی بردیم که این دو مدل دارای تغییرات مختلف رفتاری و پاتولوژیکی هیپوکامپوس هستندو فقط چند ویژگی در بین آنها مشترک هستند.هر دو مدل دارای IMT طولانی در مقایسه با گروه کنترل هستند، اما WGR فقط برای گروه CUMS کاهش داشته است و مدل LiCl-فیلوکارپین در برگیرنده رفتارهای فعالتر در مقایسه با موش های کنترل است.در هر دو مدل افت نرون هیپوکامپوس مشاهده می شود.به هر حال، فعالسازی گلیوسیز و میکروگلیا در مدل LiCl-فیلوکارپین بیشتر از مدل CUMS است و این مطلب دلالت بر این دارد که فرایند تعدیل سلولی اینفلام می تواند در افسردگی مربوط به صرع بیشتر از افسردگی اولیه مشاهده شود.

Abstract

Purpose Comorbid depression is common in patients with epilepsy. However, the epilepsy-associated depression is generally atypical and has not been fully recognized by neurologists. This study aimed to compare the behavioral and pathological changes between the chronic lithium chloride-pilocarpine rat epilepsy model (Licl-pilocarpine model) and the Chronic Unpredictable Mild Stress rat depression model (CUMS model), to evaluate for differences between epilepsy-associated depression and primary depression.

Methods The Licl-pilocarpine model and the CUMS model were established respectively and simultaneously. Spontaneous seizures were recorded by video monitoring. Forced swim test (FST) and sucrose consumption test (SCT) were performed to test depressive behaviors. Immobility time (IMT) and climbing time (CMT) in FST, sucrose preference rate (SPR) in SCT, and weight gain rate (WGR) were adopted to represent severity of depressive behaviors in rats. Immunofluorescent staining was conducted to measure expressions of neuronal specific nuclear protein (NeuN), glial fibrillary acidic protein (GFAP), and cluster of differentiation molecule 11b (CD11b) in the hippocampus of Licl-pilocarpine model, CUMS model, and Control group.

Results Significantly, more prolonged IMT was observed in both the Licl-pilocarpine model (p < 0.05) and the CUMS model (p < 0.01) than Control group. But decreased WGR was only seen in the CUMS model. The percentage of rats with CMT greater than 100 s was significantly higher in the Licl-pilocarpine model than the CUMS model (p < 0.05). Increased CMT was observed in the Licl-pilocarpine model with mild depression subgroup (EMD, IMT ≤ 100 s) even compared with the Control group. Neuronal loss was both found in the Licl-pilocarpine model and the CUMS model when comparing with the Control group (p < 0.05). However, the number of GFAP and CD11b staining cells was both greater in the Licl-pilocarpine model than the CUMS model and the Control group (p < 0.05).

Conclusion There were some different depressive behavioral and hippocampal pathological changes between the Licl-pilocarpine and the CUMS models except for some common features. Gliosis and microglial activation might be more involved in the pathophysiology of epilepsy-associated depression than primary depression.

5. Conclusions

In this study, we used Licl-pilocarpine model and CUMS model to represent epilepsy-associated depression model and primary depression model respectively, and we found that these two models had some different depressive behavioral and hippocampal pathological changes except for some common features. They both had prolonged IMT compared with Control group, but the decreased WGR was only seen in CUMS model, and Licl-pilocarpine model seemed to have more active behaviors even than Control rats. These two models both had hippocampal neuronal loss. However, more prominent gliosis and microglial activation were found in Licl-pilocarpine model than in CUMS model, indicating that gliosis and microglial cell-mediated inflam- matory process might be more greatly involved in epilepsy-associated depression than primary depression.

چکیده

ما تاثیرات تحریک های عصبی حلقه بسته و حلقه باز  بر روی توقف صرع های خود به خودی در مدل موشی صرع در هیپوکامپ موش را با استفاده از یک دستگاه تحریک عصبی درمانی برنامه پذیر کم توان سفارشی  ،  ارزیابی و مقایسه می کنیم. صرع  مزمن با استفاده از تزریق اسید کاینیک درون صفاقی در نمونه ها ایجاد شد. دو الکترود دو قطعی نیز داخل منطقه CA1 هر دو هیپوکامپ کاشته شد. این الکترود ها به دستگاه های تحریک عصبی درمانی برنامه پذیر سفارشی متصل شدند که می تواند یک تحریک عصبی به صورت دوره ای و یا بر اساس شناسایی شروع صرع بر اساس الکتروانسفالوگرافی های درون جمجمه ای (icEEE) ایجاد کند. این دستگاه شامل یک میکروچیپ متشکل از سیستم ثبت icEEG 256 کانال  و یک محرک 64 کانال و یک دستگاه شناسایی صرع برنامه پذیر می باشد که به صورت یک آرایش گیت با قابلیت برنامه پذیری میدانی (FPGA) ساخته شده است.  این محرک عصبی برای ارزیابی کارایی توقف صرع در 10 موش مبتلا به صرع ، برای مدت زمان 240 سوژه – روز (5760 سوژه – ساعت) مورد استفاده قرار گرفت. به این هدف، تمام موش ها به صورت رندوم در دو گروه قرار گرفتند. : گروه بدون تحریک و گروه همراه تحریک . گروه بدون تحریک هیچ گونه تحریکی دریافت نکرد. گروه تحریکی نیز نخست تحریک های حلقه بسته و سپس تحریک های حلقه باز را دریافت کرد. گروه های بدون تحریک و تحریک دارای خط مبنای فرکانس صرع مشابه بودند و هر روز به صورت میانگین 5 صرع را تجربه می کردند. تحریک های حلقه بسته باعث کاهش میزان تکرار صرع به مقدار 90% شد و تحریک های حلقه باز نیز میزان تکرار صرع ها را به مقدار 17% در مقایسه گروه تحریک و گروه بدون تحریک، کاهش داد.

6. جمع بندی

این مطالعه ، نشان دهنده ابعاد مهندسی مرتبط با کنترل تشنج صرعی موثر می باشد و یک دستگاه تحریک عصبی درمانی برای درمان صرع مقاوم را ارائه می کند. نتایج این مطالعه با استفاده از یک دستگاه تحریک عصبی درمانی سفارشی نشان می دهد که با استفاده از یک روش تحریک حلقه بسته می توان به صورت محسوس میزان تشنج ها را کاهش داد ( کاهش 90% در تعداد تشنج ها) ، در حالی که سیستم های حلقه باز باعث کاهش 17% می شوند. این کارایی توقف تشنج در نتیجه ایجاد یک تحریک درست قبل از شکل گیری  تشنج می باشد که در واکنش به شناسایی الگوهای غیر عادی از icEEG شکل می گیرد ، در حالی که سیستم حلقه باز تحریک ها را به صورت دوره ای ایجاد می کند. ازین رو، استراتژی های حلقه بسته باعث افزایش کارایی تحریک ها شده و در عین حال، عوارض جانبی تحریک ها را هم کاهش می دهد زیرا از کمترین تعداد تحریک ممکن استفاده می کند. ازین رو یکی از روش های جایگزین مناسب برای تحریک کننده های عصبی حلقه باز، استفاده از سیستم های حلقه بسته می باشد که در آن ها ، میزان تحریک های عمیق مغزی به کمترین تعداد ممکن می رسد. به عنوان توسعه کار های موجود در رابطه با صرع، حوزه جدید استراتژی های تحریک عمیق مغزی مبتنی بر سیستم های حلقه بسته را می توان بررسی کرد تا ابعاد مختلف آسیب شناسی فعالیت های مغزی برای درمان بیماری های مختلف عصبی را کشف نمود.

Abstract

We assess and compare the effects of both closedloop and open-loop neurostimulation of the rat hippocampus by means of a custom low-power programmable therapeutic neurostimulation device on the suppression of spontaneous seizures in a rodent model of epilepsy. Chronic seizures were induced by intraperitoneal kainic acid injection. Two bipolar electrodes were implanted into the CA1 regions of both hippocampi. The electrodes were connected to the custom-built programmable therapeutic neurostimulation device that can trigger an electrical stimulation either in a periodic manner or upon detection of the intracerebral electroencephalographic (icEEE) seizure onset. This device includes a microchip consisting of a 256-channel icEEG recording system and a 64-channel stimulator, and a programmable seizure detector implemented in a field-programmable gate array (FPGA). The neurostimulator was used to evaluate seizure suppression efficacy in 10 epileptic rats for a total of 240 subject-days (5760 subject-hours). For this purpose, all rats were randomly divided into two groups: the nostimulation group and the stimulation group. The no-stimulation group did not receive stimulation. The stimulation group received, first, closed-loop stimulation and, next, open-loop stimulation. The no-stimulation and stimulation groups had a similar seizure frequency baseline, averaging five seizures per day. Closed-loop stimulation reduced seizure frequency by 90% and open-loop stimulation reduced seizure frequency by 17%, both in the stimulation group as compared to the no-stimulation group.

VI. CONCLUSION

This study demonstrates engineering aspects associated with the effective epileptic seizure control and proposes a therapeutic neurostimulation device for the treatment of refractory epilepsy. The results of this study using a custombuilt therapeutic neurostimulation device reveal a greater efficiency at reducing ictal events (90% seizure frequency suppression) using a closed-loop stimulation method, whereas the open-loop resulted in a 17% reduction. This seizure suppression efficacy is achieved as a result of triggering a stimulation just before a seizure development, in response to an abnormal icEEG pattern detection; whereas the open-loop system delivers stimulations periodically. Thus, the closedloop strategy increases efficiency of the stimulations, while reducing the possible side effects using the minimum number of stimulations as required. Therefore, an effective alternative to the open-loop neurostimulator is the closedloop neurostimulator, in which the involvement of the deep brain stimulation is minimal. As an extension of the work on epilepsy, the new era of deep brain stimulation strategies based on closed-loop paradigms may be able to target different pathological aspects of brain activity for the treatment of various neurological disorders.
